Genetic insights into pepper-bark tree (Warburgia salutaris) reproduction in South Africa

Abstract Genetic diversity in over-exploited and threatened species has important, but often neglected, implications for conservation. In South Africa, the bark of regionally endangered pepper-bark tree, Warburgia salutaris , is extensively harvested subsistence medicine. We developed 11 polymorphic microsatellite markers to examine genetic structure among 227 W. individuals sampled from across much its range Africa’s Kruger National Park (KNP) 31 trees growing local nurseries. also tested cross-amplification success these two closely related species, elongata (IUCN listed as Endangered) stuhlmannii Vulnerable). Contrary our expectations, we found evidence clonality high all wild KNP sub-populations . identified between 114 149 unique multi-locus genotypes 258 included analyses. Bayesian assignment tests three distinct clusters that are represented by sub-population with minimal geographic some admixture. Three were individuals, i.e., nursery trees. Nursery germplasm comprised similar levels observed heterozygosity population. High reported self-incompatibility, along short distance pollination, likely explain lack recent sexual recruitment (i.e., reduced fruit set seedlings) These findings will be useful identify where genets may maximize within subpopulations restoration throughout KNP.
